Specimen Requirements

Formalin-fixed, paraffin-embedded tissue blocks; fixed in 10% neutral-buffered formalin for at least 6 hours, and not more than 72 hours. Tissue should be prepared in sections at 4 microns thick. Sections should be mounted on the positive side of an organosilane-coated slide in order to minimize detachment of the tissue from the slide during FISH assay.

Diagnostic Utility

This FISH assay detects common chromosome abnormalities and assists in the diagnosis and classification of various B-cell lymphomas.

Clinical Significance

Aggressive B-cell lymphomas are mature B-cell neoplasms associated with adverse clinical outcomes and distinct genetic alterations. The World Health Organization (WHO) Classification of Tumours of Haematolymphoid Tissues defines subsets of Diffuse large B-cell lymphoma (DLBCL) and High-grade B-cell lymphoma (HGBL) characterized by rearrangements involving MYC, MYC-IGH, BCL2, and/or BCL6. Identification of these rearrangements is critical for accurate diagnosis, prognostic stratification, and therapeutic decision-making.

Cause for Rejection

Specimen containers without two (2) patient identifiers. Specimens fixed in fixatives other than formalin are not acceptable. Acid decalcification compromises the specimen quality. Exposure to acids, strong bases or extreme heat should be avoided.
