Specimen Requirements

Formalin-fixed, paraffin embedded tissue block or cell block, or 1 unstained charged slide for each test requested plus 3-4 additional unstained slides cut at 4 microns. Adequate space on each slide for control tissue is required.

Diagnostic Utility

p57 IHC is utilized in the differential diagnosis of molar pregnancies. Complete hydatidiform moles lack nuclear staining for p57 in villous stromal cells and cytotrophoblast. Partial hydatidiform moles and non-molar chorionic villi retain expression of p57 and will reflex to Ploidy FISH analysis in this panel.

Storage and Transportation

Room temperature or refrigerated. Ship in cooled container during summer months.Ambient temperature (20-25oC; refrigerated samples acceptable); exposure to acids, strong bases or extreme heat should be avoided.

Cause for Rejection

Tissue not verified for the presence of abnormal villi and specimens fixed in fixatives other than formalin are not acceptable. Surgical pathology report should be included. Decalcification compromises the specimen quality but FISH is still attempted.

Retention

7 days.

Comments

Only tissue that contains abnormal chorionic villi (established by histopathologic criteria) should be tested. Selection of tissue for the FISH assay should be performed by a pathologist.
